E85 is a great alternative fuel source however it burns slower and cooler so does not make as much power as reg unleaded race fuel. It's almost the same principle as methanol injection to keep burn temps down in order to run more boost with less danger of detonation. Personally I would not reccomend running it alone and yes you would need a completely different tune for E85 based on my limited knowledge of it
